Monkeypox virus is an enveloped double-stranded DNA virus belonging to the Orthopoxvirus genus within the Poxviridae family. Transmission may occur through contact with bodily fluids, skin lesions, or mucosal surfaces.

Two genetically distinct clades of monkeypox virus have been described: the Central African (Congo Basin) clade and the West African clade. Historical observations have suggested differences in disease severity between these clades, with the Central African clade associated with more severe outcomes in reported outbreaks.

The BioPerfectus Monkeypox Virus Genotyping Real Time PCR Kit is a real-time PCR assay designed for the detection and differentiation of nucleic acid from the West African clade and the Central African (Congo Basin) clade of monkeypox virus. The assay targets the G2R_WA gene region for the West African clade and the C3L gene region for the Central African clade.

A housekeeping gene (RNase P) is included as an internal control for monitoring specimen quality and nucleic acid extraction.

The assay is compatible with a variety of specimen types, including tonsillar swabs, nasopharyngeal swabs, serum, whole blood, lesion exudate, and scab samples.
